Insulin Assays

Eleanor Kellon, VMD

An important issue has come up with a few private consults lately and I want to be sure to mention it here too.

Red blood cells contain an enzyme that will degrade insulin. Any amount of hemolysis in the sample for insulin determination will lower the result.  Even slight hemolysis could drop the result by about 20% and the more hemolysis there is, the greater the effect.

Spinning the sample and removing the plasma or serum ASAP will limit the effect but since most hemolysis occurs during the collection of the blood it can still be a significant problem.  Once the  cells are lysed (hemolysis) the enzyme is free in the plasma or serum.

Keeping the sample at refrigerator temperature (4 C/40 F) reduces the insulin destruction but not completely and any rewarming during shipping and handling ramps it up again.

Effect of Hemolysis on the Concentration of Insulin in Serum Determined by RIA and IRMA

Thanks Lavinia, I will go ahead & order the MovEase for Luxy & order J-herb for Spencer.  Sadly, I just opened a new bottle of Isox & hope I can return the other unopened I had bought at the same time just over a week ago, that's the way it goes!  So from what I had read if Luxy won't eat the MovEase I can use it for Spencer with the J-herb.  I'll try to post pics on.

Darcy is now up to the full dose of pergolide. She remains energetic, and her interest in food remains unchanged. Which is kind of the problem. For the past few years, she has had transient disinterest in her feed. She does not consistently finish her meals part of the time, which means she's not getting all her supplements. I understand that pergolide might help that. How long would it be before a change in feed interest might occur?

Also, right now, she's eating mostly soaked beet pulp with a handful or so of TC Lite added. She seems to like the TC Lite better. Would there be anything wrong with soaking TC Lite and using that as a carrier for her supplements and leaving off the beet pulp?

Thanks for the help,

Re: J-herb & navicular

Lavinia Fiscaletti

Hi Julia,

You might consider using Jiaogulan instead of the Isoxuprine for circulation improvement. Here is an excerpt from a previous post from Dr. Clougher:

"Jiagulan is a better circulation booster than isoxuprine, as isoxuprine doesn't last in the system long enough, and results have been equivocal. (possibly due to poor oral bioavailability and short half-life)  However, Jiaogulan shouldn't be used during the acute phase of laminitis."

Know you aren't dealing with laminitis in this case so that's not an issue. Trim is also going to be a huge factor so you are welcome to post pix on ECHoof (because Spencer is not IR/PPPID) if you'd like some input on that aspect.

Thanks, Martha - that's sort of what I thought too, but wanted to see if any of the group had experience.

re: expensive gadget - probably why the owner of the TheraPlate is charging people $150 a month for 12 sessions, trying to recoup the money spent.

Appreciate the insight!

Re: J-herb & navicular

jamsler707@...

Thanks Maggie.  Spencer was tested using Cornell for Cushings & IR (the 4 group recommended tests) in Nov. & he is definitely not either.  But last year the vet used some sort of pressure tester on his hooves & symptoms, normal pulses, laying down in his stall, which he had never done before, during the about 30 min. that he is in a stall(out on pasture & run in shed other than eat or be saddled), off, soreish when ridden.  And being on the Isox he showed no more signs.  I had quit giving it when I ran out & wasn't sure if he needed it anymore.  So when he laid down in his stall & walked not quite as normal I started it again.  I had sent Spencer's Cornell results to Jaini Clougher & she said his numbers were "beautifully normal".

So after reading all the info I guess I'll keep up the Isox & consider Phyto-Quench later if he does get sore.

Thanks again,

WasRe: J-herb & navicular Now Isox & MoveEase

jamsler707@...

I want to order some MovEase for Luxy who is Cushings, but wondered if MoveEase is compatible with Isoxsuprene for Spencer since there is a darn good chance Luxy will just leave it & I didn't know if maybe it might make Spencer more comfortable, too.  I saw on posts I found about MoveEase was compatible with Pergolide which Luxy is on, but didn't find any about Isox & MoveEase.

Thank you, I begin to think there may be hope for P. If all he was able to do was to be comfortable while being a companion to his stall mate and me, we'd all be happy. I didn't realize that there was much of a difference between the two meds. Certainly Previcox is worth a try.

Take a few minutes to search online for best price.  They vary widely.  Santa Cruz Animal Health is currently selling 60 for $108 with free shipping.

I would guess that's what your vet is selling Prascend (name brand Pergolide) for and that is a bit pricy, but unfortunately not totally out of line with the current going rate.  If the vet is willing to send in a prescription elsewhere you can find Prascend for slightly less, but I have to emphasize 'slightly' as the best price I've found without a discount coupon is $115 for 60ct (that was at www.californiapetpharmacy.com)

I am desperately looking for a reputable company selling pergolide at a
reasonable price! My vet charges $130 for 60 tablets. To me, that seems
unreasonable, but I'm also concerned about purchasing it elsewhere due
to the fact that not all pergolide is as pure as it should be.

such. I ordered mine from Valley Vet.

You can get Prascend on line. Here is a link to one source.

There are lots that use compounded pergolide from good sources.  I think they are in the group database.

Pergolide

Animal Crackers

I am desperately looking for a reputable company selling pergolide at a reasonable price! My vet charges $130 for 60 tablets. To me, that seems unreasonable, but I'm also concerned about purchasing it elsewhere due to the fact that not all pergolide is as pure as it should be.
